The accident at Yarragon was one of three involving overturned trucks on the state's roads in the space of two hours today.
Police said the milk tanker was driving along Yarragon Shady Creek Road, about 20 kilometres west of Moe, when it flipped over about 10.30am.
Victoria Ambulance spokesman John Mullen said the driver, a man in his 50s, died at the scene.
"The tanker has rolled a number of times and hit some substantial trees and the driver, the only occupant, was deceased at the scene," he said.
A Victoria Police spokeswoman said the cause of the accident was not yet known.
In a separate accident, a garbage truck flipped on its side on the Western Ring Road at Keilor Park this morning, spilling garbage across the road.
The accident occurred just before 9am and resulted in severe traffic delays in the area.
The 39-year-old driver and his female passenger, 19, were able to climb out through the shattered windscreen, Victoria Ambulance spokeswoman Susannah Wilson said.
They suffered cuts and grazes and were both taken to the Royal Melbourne Hospital in stable conditions.
Minutes earlier, another truck overturned on the Calder Freeway at Keilor Lodge, police said.
No one was seriously injured in the crash.
The accidents had been cleared and traffic was now flowing smoothly on The Western Ring Road and Calder Freeway, a VicRoads spokeswoman said.
