The rail line between Dimboola and Rainbow will be able to accommodate up to two return services per day once upgrade works are complete.
The state government has announced work has begun to replace nearly 39,000 sleepers on the line.
A government spokesman said the upgrades would take a few months, and that the line would remain open as the work will take place around scheduled train services.
The line was closed for nine months until April of this year due to safety concerns. The work to reopen the line involved replacing 5000 sleepers.

"Since reopening, 38 return freight services have used the line to transport more than 100,000 tonnes of grain, taking about 5700 trucks off our roads," the spokesman said.
Albacutya farmer Adam Gould, who farm just north of the end of the line, said he would still be using road freight to transport his yield to packing and storage sites.
"We had to shift the siding line where it comes into the GrainCorp storage silo on our property to allow access for the trucks to get in there in the interim, with VicTrack's approval" he said.
"We've also been told the Rainbow to Yaapeet line is closed, so even if I wanted to put it on a train I'd have to take it to Rainbow."
